    Das Entstehen boesartiger Tumore unter der erforderlichen immunsuppressiven Therapie stellt eines der Hauptprobleme nach Organtransplantationen dar. Wahrend durch Lebertransplantation (LTx) in den Fruhstadien des hepatozellularen Karzinoms (HCC) ein Funfjahresuberleben von 75% erreicht werden kann, sehen sich Patienten mit fortgeschritteneren Befunden durch Rezidivraten von bis zu 75% bedroht. Dieser Befund fuhrt meist innerhalb weniger Monate zum Tod. Das Auftreten dieser Rezidive wird durch die herkoemmliche immunsuppressive Therapie in Form von Calcineurininhibitoren und Corticosteroiden begunstigt. Rapamycin (Sirolimus(c)) hat sich als potentes Immunsuppressivum im Einsatz nach Organtransplantationen erwiesen. Daruberhinaus lassen erste Forschungsergebnisse auf inhibitorische Effekte auf das Wachstum verschiedener Tumorzellen schliessen. Diese Arbeit untersucht die Wirkung von Rapamycin alleine und in Kombination mit dem Calcineurininhibitor Tacrolimus (Prograf(c)) auf das Wachstum von HCC-Zellen. Hierdurch hoffen wir, den Grundstein fur einen neuen Therapieansatz in der immunsuppressiven Therapie nach LTx zu legen, durch den die Rezidivrate des HCC gesenkt werden koennte.
